Development of an Ultrasound-Assisted Rapid Digestion Method for the Quantitative Determination of Selected Trace Metals in Honey by AAS

The study deals with the development of a rapid digestion method for the determination of trace metal content (i.e., Fe, Zn) in honey. Six different honey samples were purchased from the local market and one of them was randomly selected for the optimization of the digestion method. Selected honey sample was digested using dry digestion, wet digestion and ultrasound-assisted digestion method. Ultrasound-assisted digestion was then further optimized by first selecting the reagents and the optimization of their respective ratios for better results. 0.5g sample was taken in the reagent vial and optimized digestion mixture consisted of HClO4, HCl, HNO3 and H2O2 was added into it. Then it was sonicated for 60 min at three temperature ranges i.e., 27°C, 45°C and 60°C. The solution was then filtered and diluted up to 25ml prior to analysis in the atomic absorption spectrophotometer. The optimized digestion method was then successfully applied for the analysis of selected trace metals in all the remaining honey samples. Results were also compared with the conventional i.e., dry and wet digestion methods and it was found that the optimized ultrasound-assisted digestion method was more rapid and gave better results than the conventional ones cited in the literature.
